Biographical Narrative

My career has always been marked by a commitment to interdisciplinary science. I took my BS in biology and chemistry from St. Ambrose College in Davenport, IA in 1978. My PhD is in microbiology, taken from the University of Minnesota (Minneapolis) in 1984, where I studied evolution of nosocomial drug resistance plasmids in hospital. I did a postdoctoral fellowship at Stanford University Department of Genetics from 1984-1990, where I studied self-transmissible, transposable DNA elements.
From 1990-2001 I worked in Pharma at Monsanto, Searle, Pharmacia and Pfizer (all Pfizer heritage companies), where I designed my first molecular device (the bacmid or “Bac-to-BacTM” system). Bacmid design used both plasmid and transposable DNA expertise, developed at Minnesota and Stanford, and was licensed to Life Technologies, Inc. and commercialized as “Bac-to-BacTM”, becoming one the main technology for Baculovirus cloning. Bac-to-BacTM also illustrated the potential for semi-biologic devices.
I supervised Searle efforts in phage display for pharmaceutical development. My group developed a phage-based technology to manipulate protein topologies (scanning circular permutagenesis or CP). In 1996, I joined the Monsanto Nanotechnology group, eventually assuming its leadership. We developed and patented novel protein chemoselective conjugation methods to make bioactive cytokine-nanoparticle bioconjugates and applied CP to the bioconjugates. We introduced the bioconjugates to mice, and studied immunology of these proto-nanotherapeutics. We organized the earliest international meeting focused on incorporation of biomolecules into nanodevices, along with the first scientific monograph on the topic (Lee and Savage, 1998, IBC Press), which has been used as a graduate nanotechnology text internationally. From 1999-2000, I developed a computational system that manipulates (using set theory) MHC class I binding motifs to design proteins and vaccines.
I came to OSU in 2001 to pursue hybrid technology (nanobiotechnology) uniting biotech and materials science. My group makes biomolecule/hard material hybrid devices for clinical biosensing, drug delivery and molecular imaging. We are currently focused on micro and nanoscale immunoFET sensing devices. We collaborate with colleagues in the OSU Department of Surgery (Division of Transplant) and Department of Electrical Engineering to realize these sensing devices for clinical use.
